How leucine activates mTOR signaling and why the threshold effect matters for muscle growth.
Of the nine essential amino acids, leucine holds a unique position. It is the primary anabolic trigger for muscle protein synthesis (MPS), the cellular process by which your body repairs and builds muscle tissue after training. While all essential amino acids are required for MPS to occur, leucine is the rate-limiting signal that initiates the process.
The mTOR connection
Leucine activates the mTOR (mechanistic target of rapamycin) signaling pathway, specifically mTORC1, which is the central regulator of MPS. Without sufficient leucine, mTOR activation is blunted, and MPS cannot proceed at optimal rates regardless of how much total protein you consume.
This is why leucine content in a protein source matters. Whey protein's superiority over plant proteins for muscle building has historically been attributed, in part, to its higher leucine content per gram. A post-workout meal that delivers adequate total protein but insufficient leucine will underperform on MPS relative to one with both adequate total protein and a high leucine fraction.
The threshold effect
Research from Dr. Donald Layman and colleagues at the University of Illinois has pointed to a leucine threshold (approximately 2–3 grams per meal) below which MPS stimulation is significantly reduced, and above which a ceiling is approached. This threshold model explains why spreading protein across multiple meals matters more than single large doses, and why leucine-enriching meals or shakes can be a meaningful strategy.
