Leucine and Velositol: How They Work Together, and the Doses That Matter

If you have searched for "leucine and velositol", you have probably noticed the two names keep appearing on the same labels. That is not a coincidence. Leucine is the amino acid that switches muscle protein synthesis on, and Velositol® is a patented ingredient designed to make that switch respond harder to the same amount of protein. This guide explains what each one does, what the human trials actually measured, the doses those trials used, and how to judge a leucine and velositol supplement before you buy one.
The short version
Leucine is the signal. Velositol® amplifies the response to that signal. In the published clinical work, 2 grams of Velositol® added to a small dose of whey protein doubled the muscle protein synthesis (MPS) response compared to the same whey on its own, and over eight weeks of training it improved strength, power and muscular endurance beyond what whey alone achieved. What leucine does
Of the nine essential amino acids, leucine is the one that initiates MPS. It activates mTORC1, the signalling complex that tells muscle cells to start assembling new protein. The other amino acids are the building material; leucine is the instruction to start building. We cover the mechanism in detail in our guide to leucine as the anabolic trigger.
Two practical points follow from that. First, there is a threshold. Work from Donald Layman's group at the University of Illinois and others points to roughly 2 to 3 grams of leucine per meal as the amount needed to trigger a full MPS response. Below it, the response is blunted no matter how much total protein you eat. Second, there is a ceiling. Once the threshold is cleared, piling on more leucine adds little. That is why the useful question is not "how much leucine is the most", but "does each protein feeding reliably clear the threshold". What Velositol® is
Velositol® is a patented complex of amylopectin (a highly branched starch) and chromium, developed by Nutrition21 and now sold by Everwell Health. In late 2025 the ingredient was renamed MyoVera™; the formulation did not change, so a label that says Velositol® and a label that says MyoVera™ are describing the same ingredient. It holds GRAS (Generally Recognized As Safe) status in the United States.
On a label it is easy to overlook, because 2 grams of a starch and chromium complex does not sound like much next to 25 grams of whey. Its job is not to supply protein. Its job is to change how the muscle responds to the protein you already take.
How Velositol® works with leucine
The mechanism runs through insulin signalling. Chromium supports insulin sensitivity, and the amylopectin fraction produces a small, fast insulin response when it is consumed with protein. Insulin on its own does not build muscle, but it is permissive: it improves amino acid uptake into muscle cells, dampens protein breakdown, and works alongside the leucine to mTORC1 signal. In effect, leucine tells the muscle to start synthesising protein, and Velositol® helps the muscle act on that instruction more strongly.
This is why the two are complementary rather than redundant. Leucine without a strong insulin signal still works, and Velositol® without leucine has nothing to amplify. Together, at the right doses, each covers what the other cannot.
What the clinical studies actually measured
Two human trials matter here, and it is worth knowing exactly what they tested.
2017, Journal of the International Society of Sports Nutrition (Ziegenfuss et al.). Ten healthy, recreationally active men and women took either 6 grams of whey protein alone or 6 grams of whey plus 2 grams of Velositol®, then performed eight sets of leg extensions at 80% of their estimated one-rep max. MPS, measured as muscle fractional synthetic rate, rose 24% above baseline with whey alone and 48% with whey plus Velositol®. That is the "doubles MPS" result you will see quoted, and the numbers behind it are that simple: same small protein dose, twice the synthesis response.
2021, Journal of Exercise and Nutrition (Ziegenfuss et al.). Thirty-five recreationally active middle-aged men trained for eight weeks while taking one of three daily supplements: 15 grams of whey, 30 grams of whey, or 15 grams of whey plus 2 grams of Velositol®. The Velositol® group showed greater gains in squat repetitions to failure, vertical jump height and power, and relative squat one-rep max than either whey group, including the group taking double the protein. Net protein balance was also significantly better in the Velositol® group at the four-week mark.
One honest caveat, because most articles skip it: both trials paired Velositol® with whey protein, not with isolated leucine. Whey's anabolic effect is driven largely by its leucine content (6 grams of whey contains roughly 0.6 grams of leucine, well below the threshold, which is part of why the amplification was so visible). So the studies support the leucine and Velositol® pairing by mechanism and by outcome, but a trial that combines Velositol® with a stand-alone leucine dose has not yet been published. We would rather you know that than repeat a claim the data do not quite make.
Why take leucine and Velositol® together
The logic follows from the studies. If a sub-threshold whey dose plus Velositol® can double MPS, then a leucine dose that clears the threshold on its own, plus Velositol®, gives you both a full signal and an amplified response to it. The doses that matter
The clinical dose of Velositol® is 2,000 mg. Both human trials used exactly that amount, and there is no published evidence for a smaller dose. Some products list Velositol® as a minor ingredient inside a "proprietary blend", which means you cannot tell whether you are getting 2 grams or 200 milligrams. If the label does not print the milligrams, assume it is under-dosed.

When to take it
The 2017 trial dosed immediately before training; the 2021 trial dosed daily with whey around sessions. The practical protocol is simple: take your leucine and Velositol® with a protein source within about 60 minutes of finishing your session, when the muscle is most receptive to amino acids. Safety and side effects
Velositol® is a small dose of starch plus chromium. The starch fraction contributes about 2 grams of carbohydrate, roughly 8 calories. The chromium in a serving of Lumo is 1,000 mcg, within the range used in clinical research; if you already take a separate chromium supplement, account for the total. Leucine at 5 grams is a normal dietary amount, comparable to what a large serving of chicken or whey provides. Neither ingredient is a stimulant, so the formula can be taken after evening sessions without affecting sleep. As with any supplement, speak to your healthcare provider first if you have a medical condition or take medication.
What to look for in a leucine and velositol supplement
Use this checklist before buying any product that advertises the pairing:
- Velositol® at 2,000 mg, printed on the label. Anything less has no clinical support.
- Leucine dose disclosed and at least 2.5 g per serving. If it is buried in an amino blend, you cannot verify it.
- No proprietary blends. Every active ingredient should show its milligrams.
- Third-party tested, made in a GMP-certified facility. The label should say so.
- Stimulant-free. Recovery ingredients do not need caffeine.
- Unflavored, if you already use a protein powder. It lets you add the stack to the shake you like instead of replacing it.
Frequently asked questions
Is Velositol® the same as MyoVera™? Yes. MyoVera™ is the new name Nutrition21 (now Everwell Health) gave the ingredient in 2025. Same complex, same 2-gram clinical dose.
Do I still need to eat protein if I take leucine and Velositol®? Yes. Leucine starts the process and Velositol® amplifies it, but the muscle still needs the full set of amino acids from food or a protein shake to build tissue. Think of the pair as improving the return on the protein you eat, not replacing it.
Can I take Velositol® with BCAAs or EAAs instead of whey? The mechanism does not depend on whey specifically; it depends on a leucine-rich amino acid signal plus the insulin response. That said, the human trials used whey, so whey and plant protein remain the best-supported partners.
Does Velositol® work with plant protein? Plant proteins are typically lower in leucine, which is exactly the situation where a guaranteed leucine dose and an MPS amplifier are most useful. Lumo's formula is vegetarian and unflavored, so it mixes into a pea or rice protein shake as easily as whey.
